Abstract

Present project demonstrates chemoreceptor mediated chemotaxis (Positive and Negative chemotaxis) by virtue of a new system – CHEMICAL GRADIENT MOTILITY AGAR (CGMA). Fluorescent strain of Pseudomonas was used as model organism. CGMA system consists of petriplates having a rectangular trough punched into Molten Agar, along its vertical diameter, called cell reservoir in which overnight grown culture is poured. Two more troughs were punched, one, on either sides of cell reservoir, such that all troughs have their centers on a common line. D-Glucose was used as chemo- attractant and mercuric chloride (HgCl2) as chemo- repellent. Both chemicals were poured into their respective troughs to create chemical gradients for each of them. Second system of arrangement replaces HgCl2 trough with antibiotic disc (Ampicillin). The present system was thus successful in demonstrating chemotactic movements of Pseudomonas which didn’t moved further after encountering HgCl2/ Ampicillin while, they showed dense swarming towards D-Glucose trough. Hence this project is of potential use in Bacteriology and Pathogen- Immunology.

The paper was also awarded First Prize in Paper Presentation Competition under the category of Animal Biotechnology and Microbiology at the same symposium.
